Fluorescence in situ hybridization (FISH) is based on complementary nature of double strands of DNA/RNA or DNA which is a macromolecule recognition technology. This technology uses probes which as designed DNA strands labelled with fluorophore-coupled nucleotides. These probes are used to hybridize onto the complementary sequences in the tissues or cells that are to be tested and then are visualized using an imaging system or a fluorescence microscope.

Initially, this technology was used to delineate genes within chromosomes as a physical mapping tool. FISH is used to detect presence or absence of specific sequences gene sequences in target cells or tissues.  This technology is highly precise, sensitive and specificity enabled with single gene level analytical resolution.

Due to such advantages, it immediately was used in diagnosis of genetic disorders such as subtelomeric rearrangements, microduplication/microdeletion syndromes and constitutional common aneuploidies. FISH tests are commonly used to diagnose hematologic and solid tumors and have fastest growing applications in cancer diagnostics. FISH tests are also used to detect infectious parasites and microbias such as malaria in human blood cells.
